ABSTRACT

OBJECTIVE@#To explore whether casticin (CAS) suppresses stemness in cancer stem-like cells (CSLCs) obtained from human cervical cancer (CCSLCs) and the underlying mechanism.@*METHODS@#Spheres from HeLa and CaSki cells were used as CCSLCs. DNA methyltransferase 1 (DNMT1) activity and mRNA levels, self-renewal capability (Nanog and Sox2), and cancer stem cell markers (CD133 and CD44), were detected by a colorimetric DNMT activity/inhibition assay kit, quantitative real-time reverse transcription-polymerase chain reaction, sphere and colony formation assays, and immunoblot, respectively. Knockdown and overexpression of DNMT1 by transfection with shRNA and cDNA, respectively, were performed to explore the mechanism for action of CAS (0, 10, 30, and 100 nmol/L).@*RESULTS@#DNMT1 activity was increased in CCSLCs compared with HeLa and CaSki cells (P<0.05). In addition, HeLa-derived CCSLCs transfected with DNMT1 shRNA showed reduced sphere and colony formation abilities, and lower CD133, CD44, Nanog and Sox2 protein expressions (P<0.05). Conversely, overexpression of DNMT1 in HeLa cells exhibited the oppositive effects. Furthermore, CAS significantly reduced DNMT1 activity and transcription levels as well as stemness in HeLa-derived CCSLCs (P<0.05). Interestingly, DNMT1 knockdown enhanced the inhibitory effect of CAS on stemness. As expected, DNMT1 overexpression reversed the inhibitory effect of CAS on stemness in HeLa cells.@*CONCLUSION@#CAS effectively inhibits stemness in CCSLCs through suppression of DNMT1 activation, suggesting that CAS acts as a promising preventive and therapeutic candidate in cervical cancer.

RESUMEN Introducción: El agua minero-medicinal estudiada procede del Complejo Termal de Presidencia Roque Sáenz Peña, provincia del Chaco, ubicado en el nordeste de Argentina. Las aguas de los acuíferos terciarios del norte argentino presentan un marcado gradiente de salinidad desde el sector oeste (subgrupo Jujuy) hasta el sector este (formación Chaco). Los acuíferos son procesos dinámicos y, en consecuencia, la composición del agua subterránea natural debe contemplarse con la perspectiva de su posible variación espacio-temporal. Objetivos: Analizar y comparar la composición química y la calidad microbiológica del agua de las termas de la ciudad de Sáenz Peña en el período 2016-2018 y evaluar sus principales características fisicoquímicas que definen sus potenciales aplicaciones terapéuticas. Resultados: El agua termal por su temperatura se clasifica como agua hipotermal, con pH cercano a la neutralidad, conductividad elevada mostrando su relación directa con la alta salinidad. Por su porcentual equivalente se define como agua clorurada, con alto contenido del anión sulfato y los cationes sodio, potasio, calcio y magnesio. De acuerdo con la composición mayoritaria de cloruros, el uso terapéutico vía tópica sería recomendado para afecciones óseas y musculares, así como problemas dermatológicos y circulatorios.


SUMMARY Introduction: The mineral-medicinal water studied comes from the Presidencia Roque Sáenz Peña Thermal Complex, Chaco province, located in the northeast of Argentina. The waters of the tertiary aquifers of northern Argentina present a marked salinity gradient from the western sector (Jujuy subgroup) to the eastern sector (Chaco formation). Aquifers are dynamic processes and consequently, the composition of natural groundwater must be viewed from the perspective of its possible spatial-temporal variation. Aims: To analyse and to compare the chemical composition and microbiological quality of thermal water of the city of Sáenz Peña in the 2016-2018 period and to evaluate its main physicochemical characteristics that define its potential therapeutic applications. Results: Due to its temperature, the thermal water is classified as hypothermal water, with a pH close to neutrality, high conductivity, showing its direct relationship with high salinity. Its equivalent percentage defined as chlorinated water, with high content of the sulphate anion and the cations sodium, potassium, calcium and magnesium. According to the majority composition of chlorides, topical use is recommended for bone and muscle ailments, as well as dermatological and circulatory problems.


RESUMO Introdução: A água mineral-medicinal estudada provém do Complexo Térmico Presidencia Roque Sáenz Peña, província de Chaco, localizada no nordeste da Argentina. As águas dos aquíferos terciários do norte da Argentina apresentam um acentuado gradiente de salinidade do setor ocidental (subgrupo Jujuy) ao setor oriental (formação Chaco). Os aquíferos são processos dinâmicos e, consequentemente, a composição da água subterrânea natural deve ser vista sob a perspectiva de sua possível variação espaço-temporal. Objetivos: Analisar e comparar a composição química e qualidade microbiológica das águas termais da cidade de Sáenz Peña no período 2016-2018 e avaliar suas principais características físico-químicas que definem seus potenciais aplicações terapêuticas. Resultados: Devido à sua temperatura, a água termal é classificada como água hipotérmica, com pH próximo da neutralidade, alta condutividade, mostrando sua relação direta com a alta salinidade. Seu percentual equivalente definido como água clorada, com alto teor do ânion sulfato e dos cátions sódio, potássio, cálcio e magnésio. De acordo com a composição majoritária dos cloretos, o uso tópico é recomendado para doenças ósseas e musculares, bem como para problemas dermatológicos e circulatórios.

ABSTRACT

Se presenta el caso clínico de un adolescente de 18 años de edad, atendido en el Policlínico Docente Armando García Aspurú de Santiago de Cuba por presentar antecedentes de retardo en el desarrollo psicomotor, marcha anadeante, caídas frecuentes, signo de Gowers positivo, fuerza muscular proximal disminuida en miembros superiores e inferiores, pseudohipertrofia de los gemelos, atrofia de cuádriceps pectoral y escapular. Los signos y síntomas clínicos permitieron diagnosticar una distrofia muscular de Duchenne. Como no pudo realizarse el tratamiento rehabilitador necesario, se produjo un deterioro músculo - esquelético progresivo y severo del paciente.


The case report of an 18 years adolescent is presented. He was assisted at Armando García Aspurú Teaching Polyclinic in Santiago de Cuba due to a history of psychomotor development retardation, wandering march, frequent falls, positive Gowers sign, diminished proximal muscular force in upper and lower limbs, twins pseudohypertrophy, atrophy of pectoralis and scapular quadriceps. The clinical signs and symptoms allowed to diagnose a Duchenne muscular dystrophy. As the necessary rehabilitative treatment could not be carried out, a progressive and severe musculoskeletal deterioration of the patient took place.

ABSTRACT

O tema é como é vista a ação terapêutica em psicanálise hoje e no passado. Defendo a ideia de que se assiste com frequência a uma sobrevalorização das visões e intervenções tradicionais com desconhecimento das conceptualizações e descobertas mais recentes, e noutros casos por uma preferência pelas novas formas de conceptualização e intervenção e desvalorização excessiva das mais antigas. Conceitos-chaves no processo terapêutico como a recordação, o insight, a interpretação, a resistência, neutralidade e a transferência foram reformulados ou deixaram de ser o centro da atuação e viram diminuída a sua importância, cedendo a relevância a conceitos como a contratransferência, identificação projetiva e, mais recentemente, a relação, o enactment, a emoção, as relações interpessoais fora do setting, e o futuro do paciente. A psicanálise virou a sua atenção do paciente para analista. Discutem-se as mudanças das visões tradicionais em visões contemporâneas e as suas implicações em dois textos. Neste texto, identificado como a primeira parte, vamos abordar a recordação, o insight, interpretação, resistência e neutralidade. Os restantes serão abordados na segunda parte.


The topic is how the therapeutic action in psychoanalysis is seen today and in the past. I support the idea that there is often an overvaluation of traditional views and interventions with ignorance of the latest conceptualizations and discoveries and in some cases a preference for new forms of conceptualization and intervention and excessive devaluation of the older ones. Key concepts in the therapeutic process such as recall, insight, interpretation, resistance, neutrality, and transference have been reformulated or ceased to be the center of action and have diminished their importance, yielding relevance to concepts such as countertransference, projective identification, and more recently to relation, enactment, emotions, interpersonal relationships outside the setting, and the patient's future. Psychoanalysis turned its attention from the patient to the analyst. We discuss the changes of traditional visions in contemporary visions and their implications in two texts. In this text identified as the first part we will address recall, insight, interpretation, resistance, and neutrality. The remainder will be covered in Part Two.


El tema es cómo se ve la acción terapéutica en el psicoanálisis hoy y en el pasado. Apoyo la idea de que a menudo hay una sobrevaloración de los puntos de vista e intervenciones tradicionales con ignorancia de las últimas conceptualizaciones y descubrimientos y, en algunos casos, una preferencia por nuevas formas de conceptualización e intervención y una devaluación excesiva de las más antiguas. Los conceptos clave en el proceso terapéutico, como el recuerdo, el insight, la interpretación, la resistencia, la neutralidad y la transferencia, se han reformulado o dejado de ser el centro de acción y han disminuido su importancia, dando relevancia a conceptos como la contratransferencia, la identificación proyectiva y más recientemente, a relación, el enactment, emociones, relaciones interpersonales fuera del entorno y el futuro del paciente. El psicoanálisis dirigió su atención del paciente al analista. Discutimos los cambios de las visiones tradicionales en las visiones contemporáneas y sus implicaciones en dos textos. En este texto identificado como la primera parte, abordaremos el recuerdo, el insight, la interpretación, la resistencia y la neutralidad. El resto se cubrirá en la Parte Dos.

ABSTRACT

Aim To determine whether 8-bromo-7-me-thoxychrysin ( BrMC) inhibits in vitro carcinogenicity via up-regulating miR-519d expression and down-regu-lating Twist1 expression in liver cancer stem-like cells ( LCSLCs) derived from SMMC-7721 cell line. Meth-ods The second generation spheroids derived from SMMC-7721 cell line were obtained by sphere-forming assay and were considered as LCSLCs . Then LCSLCs were treated with various concentrations ( 1.0, 3.0, 10.0 μmol·L-1) of BrMC. The expression level of miR-519d was detected using real-time PCR. And in vitro carcinogenicity was investigated by sphere-forming assay and clone-forming assay in agar. The transcrip-tional activity and protein expression of Twist1 were an-alyzed using luciferase reporter assay and Western blot. Moreover, the molecular mechanism of BrMC was elucidated via miR-519 mimic transfection and Twist1 gene transduction, respectively. Results Compared with SMMC-7721 cells, miR-519d-3p was low-ex-pressed and Twist1 was over expressed in LCSLCs. And the sphere-forming ratio and the clone-forming ra-tio decreased by treatment with BrMC ( 1.0, 3.0, 10.0 μmol·L-1) in a dose-dependent manner. Fur-thermore, luciferase reporter assay demonstrated miR-519d could directly target the 3′ untranslated region of Twist1 mRNA and regulate protein expression. miR-519d mimic enhanced the effects of BrMC (3.0 μmol ·L-1) . However, Twist1 gene transduction effective-ly reversed the effects of BrMC ( 3.0 μmol·L-1) . Conclusion BrMC inhibits in vivo carcinogenicity via regulating miR-519/Twist1 signal axis in LCSLCs de-rived from SMMC-7721 cell line.

ABSTRACT

Objective To observe the effect of a Uyghur medicine Munziq Balgam on the mouse model of psoriasis.Methods 130 healthy Kunming mice were used in this study .The mouse model of psoriasis was established by applying 5% propranol hydrochloride emulsion to the mouse ear .The tail and ear skin histology was examined using HE staining, and the serum levels of IL-10, IL-17, IL-23 were measured with ELISA.Results The mice of Munziq Balgam treated groups (2, 4, 8 g/kg) did not show obvious abnormalities of general condition and body weight changes (P >0.05).The 8 g/kg Munziq Balgam treated group showed decreased thymus index (P 0.05). The 2, 4, and 8 g/kg Munziq Balgam treated groups showed promoted formation of epidermis granular layer in the mouse tail skin and improved histology of the ear skin .Conclusions The Uygur medicine Munziq Balgam shows therapeutic effect on experimental mouse models of psoriasis .

ABSTRACT

O fazer psicanalítico em suas variedades será o tema deste trabalho. Inicialmente, a situação analisante será objeto de um exame para identificar sua estrutura e dinâmica. Em seguida, serão consideradas algumas especificidades da situação analisante em diferentes condições da prática psicanalítica, para mostrar que em todas as suas variações o manejo da situação analisante é base de tudo o que de mais específico se possa ou deva aí fazer, revelando-se também uma eficácia terapêutica própria ao dispositivo.


This paper is about the practice of psychoanalysis in its manifold forms. The first part is about the analyzing situation and the analytic setting: their structure and dynamism are dealt with in a panoramic and historic view. The point here is to show that the main task of the analyst should always be to establish and to maintain the analyzing situation in its most lively and dynamic form. The second part is about specific aspects of the analyzing situation under different conditions of the psychoanalytic practice, and aims to show that the managing (or holding) of the analyzing situation is, in all cases, the basic and most important aspect of all the other activities that can be used; in fact it is good managing of the analyzing situation that best reveals its therapeutic efficacy.


La práctica psicoanalítica en sus variantes será el objeto de este trabajo. Inicialmente, la situación en análisis será objeto de un examen para determinar su estructura y su dinámica. A continuación se considerarán algunos aspectos específicos de la situación en análisis en diferentes condiciones de la práctica psicoanalítica, para demostrar que en todas sus variantes la gestión de la situación de análisis es la base de todo lo específico que se pueda o deba hacer, revelándose también una eficacia terapéutica propia del dispositivo.

ABSTRACT

Tablet 912 - II. active ingredients from Fructus Tribuli, was applied for treating 40 cases of cerebral vascular disorders. After a course of 3 months, it was found that the tablet markedly improved the clincal symptoms, lowered the blood viscosity, regulated platelet functions, ameliorated the microcirculation, increased the cerebral blood flow. When compared with Carlan tablet, the action of improving clinical manifestations was basically the same, while for that of correlated clinical indices, 912-II is better than the latter tablet.
